The Berryman Trail in the Ozarks of Missouri is about 3 hrs SW of St. Louis. Have never been there but it is on my to do list of trails. Bonkfest was held their a month ago. Check mtbr for more info.

ohmygodyouMUST do Berryman if you're that close. Right there, you have a choice of 75+ miles of sweet, sinuous, salivation inducing singletrack to ride. Berryman offers 25 miles, Council Bluffs also has about 25, and there's a connector trail that I think is also 25 miles. ALL of it Single track. ALL of it really, really good. I was down there three years ago, and I need to get back there soon. VERY soon. Of all the trails I've ridden(plenty), Berryman is way up on the list of the best.

I've heard nothing but great stuff about the Berryman trail. I was going to ride it this fall but plans got changed -BUMMER! Anyway, in doing research on the area and other trails, I came across this site: http://www.stlbiking.com/Menu_Trails.htm It's got a lot of info on St. Louis and the surrounding area. Definitely worth a look. Castlewood and the Chubb trail are supposed to be pretty good.
